چکیده

Results Strong evidence for association was seen for both STAT4 (strongest effect rs7574865 OR 1.24 95%CI 1.1–1.4, ptrend = 0.0008) and TRAF1/C5 (strongest effect rs2900180 OR 1.23 95%CI 1.1–1.38, ptrend = 0.0004). The 6q23 region showed weak evidence for association with JIA (rs6920220 OR 1. 17 95% CI 1.01–1.36, ptrend = 0.03). In all cases the associated allele was the same as for RA and the effect sizes were similar.

Overlap of disease susceptibility loci for rheumatoid arthritis and juvenile idiopathic arthritis

BACKGROUND Genome-wide association studies (GWAS) have been extremely successful in the search for susceptibility risk factors for complex genetic autoimmune diseases. As more studies are published, evidence is emerging of considerable overlap of loci between these diseases. In juvenile idiopathic arthritis (JIA), another complex genetic autoimmune disease, the strategy of using information fro...

The TRAF1/C5 region is a risk factor for polyarthritis in juvenile idiopathic arthritis

OBJECTIVE Juvenile idiopathic arthritis (JIA) is a chronic disorder in which both genetic and environmental factors are involved. Recently, we identified the TRAF1/C5 region (located on chromosome 9q33-34) as a risk factor for rheumatoid arthritis (RA) (p(combined) = 1.4 x 10(-8)). In the present study the association of the TRAF1/C5 region with the susceptibility to JIA was investigated. MET...

Re-evaluation of putative rheumatoid arthritis susceptibility genes in the post-genome wide association study era and hypothesis of a key pathway underlying susceptibility

Rheumatoid arthritis (RA) is an archetypal, common, complex autoimmune disease with both genetic and environmental contributions to disease aetiology. Two novel RA susceptibility loci have been reported from recent genome-wide and candidate gene association studies. We, therefore, investigated the evidence for association of the STAT4 and TRAF1/C5 loci with RA using imputed data from the Wellco...

Evaluation of the rheumatoid arthritis susceptibility loci HLA-DRB1, PTPN22, OLIG3/TNFAIP3, STAT4 and TRAF1/C5 in an inception cohort

INTRODUCTION This study investigated five confirmed rheumatoid arthritis (RA) susceptibility genes/loci (HLA-DRB1, PTPN22, STAT4, OLIG3/TNFAIP3 and TRAF1/C5) for association with susceptibility and severity in an inception cohort. METHODS The magnitude of association for each genotype was assessed in 1,046 RA subjects from the Yorkshire Early RA cohort and in 5,968 healthy UK controls. Additi...

Genetic risk factors for rheumatoid arthritis differ in Caucasian and Korean populations.

OBJECTIVE Recent studies have identified a number of novel rheumatoid arthritis (RA) susceptibility loci in Caucasian populations. The aim of this study was to determine whether the genetic variants at 4q27, 6q23, CCL21, TRAF1/C5, and CD40 identified in Caucasians are also associated with RA in a Korean case-control collection. We also comprehensively evaluated the genetic variation within PTPN...
